aquifer=20
                        systems, and particularly the role of =
prokaryotic and=20
                        eukaryotic microbes, is poorly understood.&nbsp; =
A large=20
                        body of work in marine and freshwater systems =
has=20
                        indicated that eukaryotes such as protozoa play=20
                        fundamental roles in the carbon and nutrient =
cycles in=20
                        all aquatic environments =96 ranging from =
nutrient-rich=20
                        coastal systems to nutrient-poor mid-ocean =
gyres.&nbsp;=20
                        Nevertheless, the role of protozoa in aquifer =
systems=20
                        has been examined by a relatively small number =
of=20
                        studies.&nbsp; Given the pivotal role these =
organisms=20
                        play in other aquatic environments, it is =
reasonable to=20
                        assume that they play fundamental roles in the =
carbon=20
                        and nutrient cycles of the subsurface.&nbsp; It =
is time=20
                        to investigate which protozoan species are =
present,=20
                        which bacteria they graze upon and the impact of =

                        protozoan grazing on the carbon and nutrient =
cycles in=20
                        groundwater.&nbsp; The goal of this work is to =
utilize=20
                        stable isotopes in combination with geochemical =
and=20
                        microbial methods to study the effect of =
protozoa on=20
                        nutrient regeneration rates and carbon =
remineralization=20
                        in a subterranean estuary on Cape Cod MA.&nbsp; =
This=20
                        work will use <SUP>13</SUP>C-labeled substrates =
such as=20
                        acetate to track the active bacterial and =
protozoan=20
                        community.&nbsp; Sediment will be collected from =
the=20
                        freshwater and saline end-members of the =
subterranean=20
                        estuary and will be used to establish =
flow-through=20
                        columns.&nbsp; These columns will be incubated =
with=20
                        groundwater pumped from the appropriate site in =
the=20
                        estuary to maintain the <I>in situ</I> salinity =
and ion=20
                        composition.&nbsp; Groundwater will be amended =
with=20
                        <SUP>13</SUP>C-substrates.&nbsp; The movement of =
the=20
                        isotopic label from substrate to organic matter =
will be=20
                        monitored by isotope-ratio mass spectrometry of=20
                        inorganic carbon (aqueous CO<SUB>2</SUB> =
species), bulk=20
                        organic carbon (TOC / DOC) and small organic =
molecules=20
                        (volatile organic acids and amino acids).&nbsp;=20
                        Incorporation of the label into microbial =
biomass will=20
                        be assessed by examination of =
isotopically-labeled=20
                        nucleic acids and phospholipid fatty acids=20
                        (PLFAs).&nbsp; Active bacterial and protozoan =
species=20
                        will be identified by their isotopically-labeled =
rDNA or=20
                        rRNA sequence.&nbsp; The relative roles of =
protozoa and=20
                        bacteria will be assessed by parallel column =
incubations=20
                        with 1.0-=B5m filtered and non-filtered =
groundwater. The=20
                        proposed work is the first study, to our =
knowledge, that=20
                        directly links protozoan community composition =
to=20
                        nutrient regeneration and carbon =
remineralization in=20
                        fresh and saline aquifers.&nbsp; This group of =
microbes=20
                        is vastly under-studied and the results of this =

                        have the potential to elucidate recycling =
processes=20
                        outside the current paradigm of microbial =
activity in=20
